      Biotin is a vitamin that binds with high affinity to avidin and streptavidin. Biotin can be conjugated to many proteins without altering their biological activities due to its small size (244 Da). Biotinylated proteins can be tested or captured by streptavidin or avidin, and be applied in ELISA, dot blot or Western blot experiments. N-Hydroxysuccinimide (NHS) activated biotins are the most popular type of biotinylation reagents, which can react efficiently with primary amino groups (-NH2) to form stable amide bonds.

      Sulfosuccinimiyl Biotin (Biotin-Sulfo MHS) can enable simple and efficient biotinylation of antibodies, proteins and any other primary amine-containing macromolecules in solution.

      Biotin-Sulfo NHS Microscale Antibody Labeling Kit contains reagents sufficient for labeling 20~100μg antibodies. The whole process can be finished within 2 hours with only around 30 minutes for operating. Labeled antibodies can be separated from free dye using an ultrafiltration tube, and the recovery rate can be around 60%~90%.       Biotin-Sulfo NHS is water solubility compared to ordinary NHS-ester compounds and dissolves readily in polar solutions. It is charged by the sodium sulfoxide group on the succinimidyl ring, which makes it unable to permeate cell membranes and only able to label the surface proteins of whole cells.

      Cell surface biotinylation has emerged as an important tool for studying the expression and regulation of receptors and transporters, differentiation of plasma membrane proteins from those localized to organelle membranes, and distribution of membrane proteins in polarized epithelial cells.

Precautions

- Biotin-Sulfo NHS Microscale Antibody Labeling Kit can tolerate protein samples containing low concentrations of glycerol, Tris or merthiolate, but not those containing carriers like BSA.

- A standard Biotin-Sulfo NHS labeling process can be used for labeling 50μg IgG, with a DOL between 2 and 6. Please check the total mass of IgG for labeling before starting the process.

- The final concentration of IgG in the standard reaction system of this kit should not be lower than 1mg/mL. 

- A modified molar ratio of Biotin-Sulfo NHS /IgG is needed, if you prefer a higher DOL for your IgG.

- The 2× Biotin-Sulfo NHS Reaction Buffer in this kit has been optimized based on Biotin-Sulfo NHS. Using 2× Biotin-Sulfo NHS Reaction Buffer with other dyes is not recommended, which may affected the final DOL of your IgG.
