Product Description
Biotin is a vitamin that binds with high affinity to avidin and streptavidin. Biotin can be conjugated to many proteins without altering their biological activities due to its small size (244 Da). Biotinylated proteins can be tested or captured by streptavidin or avidin, and be applied in ELISA, dot blot or Western blot experiments. N-Hydroxysuccinimide (NHS) activated biotins are the most popular type of biotinylation reagents, which can react efficiently with primary amino groups (-NH2) to form stable amide bonds.
Biotin-PEG4 NHS ester enables simple and efficient biotin labeling of antibodies, proteins and any other primary amine‐containing macromolecule. The hydrophilic polyethylene glycol (PEG) spacer arm in Biotin-PEG4 NHS ester imparts its water solubility that is transferred to the biotinylated molecule. Consequently, antibodies that have been labeled with Biotin-PEG4-NHS ester exhibit less aggregation when stored in solution compared to antibodies labeled with reagents having only hydrocarbon spacers. The PEG spacer also provides a long and flexible linkage between the biotin and antibody which reduces the interference of steric hindrance when binds to the avidin/streptavidin. Biotin-PEG4 NHS Microscale Antibody Labeling Kit contains reagents sufficient for labeling 20~100μg antibodies. The whole process can be finished within 2 hours with only around 30 minutes for operating. Labeled antibodies can be separated from free dye using an ultrafiltration tube, and the recovery rate can be around 60%~90%. Precautions
- Biotin-PEG4 NHS Microscale Antibody Labeling Kit can tolerate protein samples containing low concentrations of glycerol, Tris or merthiolate, but not those containing carriers like BSA.
- A standard Biotin-PEG4 NHS labeling process can be used for labeling 50μg IgG, with a DOL between 2 and 6. Please check the total mass of IgG for labeling before starting the process.
- The final concentration of IgG in the standard reaction system of this kit should not be lower than 1mg/mL.
- A modified molar ratio of Biotin-PEG4 NHS/IgG is needed, if you prefer a higher DOL for your IgG.
- The 2× Biotin-PEG4 NHS Reaction Buffer in this kit has been optimized based on Biotin-PEG4 NHS. Using 2× Biotin-PEG4 NHS Reaction Buffer with other dyes is not recommended, which may affected the final DOL of your IgG.
