Biotin is a vitamin that binds with high affinity to avidin and streptavidin. Biotin can be conjugated to many proteins without altering their biological activities due to its small size (244 Da). Biotinylated proteins can be tested or captured by streptavidin or avidin, and be applied in ELISA, dot blot or Western blot experiments. N-Hydroxysuccinimide (NHS) activated biotins are the most popular type of biotinylation reagents, which can react efficiently with primary amino groups (-NH2) to form stable amide bonds.
Biotin-LC NHS, also known as Biotin-X NHS, contains a spacer arm off the valeric acid side chain of biotin with an NHS ester group at its end. Biotin-LC NHS can enable simple and efficient biotinylation of antibodies, proteins and any other primary amine-containing macromolecules in solution.
The 6-aminocaproic acid spacer (22.4 angstroms) of Biotin-LC NHS ester greatly increases the length between a covalently modified molecule and the bicyclic biotin rings leading to a better binding potential for avidin or streptavidin probes. NHS-LC-biotin is insoluble in aqueous environments requiring the dissolution of organic solvents prior to the addition to a buffered reaction.
