      Goat anti-mouse antibody can be conjugated to other proteins by thiol-maleimide and hydrazone coupling approaches. 

     Thiol-maleimide method is commonly performed with a bifunctional linker, such as the commonly used SMCC. However, SMCC-modified protein is extremely unstable and often self-reactive since proteins often contain both amine and thiol groups that cause a significant amount of homo-crosslinking. The maleimide conjugates can re-arrange, hydrolyze, or exchange in the presence of thiols/cysteine.

     In the hydrazone coupling method, the conjugation reaction is based on a reaction between hydrazine and aldehyde. This method is low yielding although a catalyst is used. The size of the molecule and inaccessibility of the functional group contribute to low yield. In addition, linkage stability is an issue.

     Antibody conjugation kit is based on a 3rd generation protein crosslink technology, which has been engineered to solves all the problems above and allow rapid and quantifiable conjugation between streptavidin and target antibody/protein. The conjugation efficiency can be raised up to 80~95%. The whole operation time can be finished in less than 1 hour.

     This kit offers the L-Linker labeled goat anti-mouse antibody. Target proteins can be labeled with S-Linkers in this kit. When the L-Linker labeled goat anti-mouse antibody and the S-Linker labeled target protein are mixed together, a rapid and quantifiable conjugation then initiates with no catalysts at all.

     This kit can be used for labeling 50~200μg proteins, and only for 1 reaction. If you need a package for a different size, please contact our sales manager.

     In this kit, S-Linkers are activated by NHS ester, which can react with the -NH2 in proteins. If you need these linkers to be with other types of esters for disulfide re-bridging or glycosylation chain labeling, please contact our sales manager.

Precautions

- The molecular weight cut-off of the 10K, 500μL ultrafiltration tubes in this kit in around 10kDa, which can be applied for proteins larger than 10kDa.

- The maximum loading volume of the 10K, 500μL ultrafiltration tubes is 500μL, and the minimum loading volume is 20μL. The retention volume can be less than 5μL.

- The ultrafiltration membrane can be teared or pierced by pipette tips, so please try not to touch the ultrafiltration membrane with the shape end of the pipette tip.

- Protein stabilizers, such as BSA, in samples may affect the final yield, so please remove these stabilizers before starting.

- Non-protein stabilizers, such as Tris or glycerol, in samples may affect the final yield, so please remove these stabilizers with the ultrafiltration tube and the 1× Dilution Buffer.

- The final concentration of target proteins in the standard reaction system of this kit should not be lower than 1mg/mL. If the initial concentration of the sample is too low, you can concentrate it with the ultrafiltration tube and the 1× Dilution Buffer, and make the concentrated sample up to over 2mg/mL.
