      Site-Specific Oligonucleotide Antibody Labeling Kit provides an optimized strategy for a stable linkage between antibodies and oligonucleotides with a high conjugation efficiency and an ultra-fast reaction speed.

      The antibody labeling strategies based on amine-NHS ester or maleimide chemistry will produce random labels. In some cases, labeling in Fab areas could block antigen binding. 

      This kit is based on a site-specific cysteine/disulfide chemistry strategy. The P-Linkers in this kit are inserted to disulfide bond location to re-form the disulfide bridging without affecting antibody-antigen binding. On the other side, the amine modified oligonucleotides can be labeled by the N-Linkers in this kit based on an amine-NHS ester strategy. Once the antibodies labeled with P-Linkers and the oligonucleotides labeled with N-Linkers are mixed together, the conjugation reaction reacts instantly.

      This kit can be used for labeling 50~500μg antibodies, and only for 1 reaction. If you need a package for a different size, please contact our sales manager.

In this kit, the degree of labeling (DOL) of the P-linker labeled antibodies can be measured by using N-Linker labeled fluorescent dyes. If you need these fluorescent dyes, please contact our sales manager.

Precautions

- This kit can be used for the conjugation between antibodies and oligos (10-120 bases long single-stranded oligos or up to 80 bases long double-stranded oligos). The target sites for labeling in the oligos should be amine modified which can be done in a regular Oligo synthesis lab.

- Protein stabilizers, such as BSA, in samples may affect the final yield, so please remove these stabilizers before starting.

- Non-protein stabilizers, such as Tris or glycerol, in samples may affect the final yield, so please remove these stabilizers with the ultrafiltration tube and the 1× Dilution Buffer.

- The final concentration of target antibodies in the standard reaction system of this kit should not be lower than 1mg/mL. If the initial concentration of the sample is too low, you can concentrate it with the ultrafiltration tube and the 1× Dilution Buffer, and make the concentrated sample up to over 2mg/mL.

- The molecular weight cut-off of the 10K, 500μL ultrafiltration tubes in this kit in around 10kDa, which can be applied for proteins larger than 10kDa.

- The maximum loading volume of the 10K, 500μL ultrafiltration tubes is 500μL, and the minimum loading volume is 20μL. The retention volume can be less than 5μL.

- The ultrafiltration membrane can be teared or pierced by pipette tips, so please try not to touch the ultrafiltration membrane with the shape end of the pipette tip.
