      AF 546 SE is the 1 amino-reactive dye that can be coupled to primary amines (R-NH2) on almost any protein, antibody, or amine-modified oligonucleotide. The AF 546 SE Micro Antibody Labeling Kit provides the 1 method for rapid labeling of micro amounts (20~100 μg) of antibody using AF 546 SE. Antibody labeling can usually be completed within 2 hours, and the manual operation time is about 30 minutes. The labeled antibody can be rapidly desalted and purified by ultrafiltration membrane, and the recovery rate is usually 60% ~ 90%.

Precautions

- The AF 546 SE Micro Antibody Labeling Kit is compatible with low concentrations of glycerol, Tris, and thimerosal, but stable proteins must be removed from the sample before labeling.

- The molar ratio of dye to antibody molecules in a standard AF 546 SE labeling step has been optimized to label 50 μg of IgG antibody with a DOL in the range of 2 to 6. Therefore, before labeling, it is necessary to confirm the amount of μg of IgG to be labeled.

- In the standard reaction system of this kit, the final concentration of IgG should not be less than 1 mg/mL. If you need a higher DOL, you may need to adjust the molar ratio of dye to antibody molecules in the reaction system.

- The 2 × AF 546 SE reaction buffer provided in this kit has been optimized according to the characteristics of AF 546 SE. Therefore, it is not recommended to mix with other activated ester micro antibody labeling kits in the same series, which may cause the final DOL to be inconsistent with expectations.
